    Genetics: Chromosome 20: ARFGEF2, CSE1L, STAU1

Looking at some of the 25 'critical SNPs' in the first PrecisionLife study on ME/CFS from, I see that one of them, rs237475 which was mapped to the gene KCNB1, is very near this most significant locus in DecodeME. This is the significance plot for DecodeME, showing the ARFGEF2 locus just a...

Should we initiate development of a new, short questionnaire to identify PEM (to aid diagnosis)?

I'm just suggesting that, even if imperfect, it makes more sense than the criteria being a mishmash of random symptoms that don't really even seem related. For example, one requirement for CCC: So you could fulfill this specific category (which to be fair is only one category of the criteria)...

Should we initiate development of a new, short questionnaire to identify PEM (to aid diagnosis)?

Sure, I think it'd also be valid to separate out people with well-characterized diseases.

Should we initiate development of a new, short questionnaire to identify PEM (to aid diagnosis)?

Oh yes sorry mixed up my specificity and sensitivity. But yes, primarily focused on making sure there are as few false positives as possible. You mean excluding people who might have another condition that explains PEM? I don't really agree with that view. If they have PEM, they have PEM...

Should we initiate development of a new, short questionnaire to identify PEM (to aid diagnosis)?

I wonder if instead of focusing on trying to create a questionnaire with optimal accuracy in separating healthy from ME/CFS, it might be better to think about making a questionnaire with virtually 100% specificity, at the cost of poor sensitivity. Meaning make a questionnaire where only people...

Preprint Initial findings from the DecodeME genome-wide association study of myalgic encephalomyelitis/chronic fatigue syndrome, 2025, DecodeMe Collaboration

Quick and potentially not totally accurate summary: They created a machine learning model designed to predict whether participants were cases or controls based on rare variants in their DNA. The prediction algorithm was connected to an external database of known protein interactions (STRING)...

Preprint Initial findings from the DecodeME genome-wide association study of myalgic encephalomyelitis/chronic fatigue syndrome, 2025, DecodeMe Collaboration

I think just much smaller studies or studies that didn't use as strict of definitions. For example, the UK BioBank has WGS data for their chronic fatigue syndrome phenotype, and it was included in a rare variant study, but the cohort is smaller than DecodeME and the definition of CFS is more...
