Coronavirus - worldwide spread and control

Message from the moderation team: With the support of the committee, the moderation team has decided to permanently close the 'Coronavirus - Worldwide Spread and Control' thread, and not allow further posting of, or discussing, data on infection and death rates around the world, and discussion...

A cohort study of whether parental separation and lack of contact [...] predicts disease severity [...] in young peoples ME/CFS, 2020, O'Donnell et al

Great points on this thread. If these people wanted to delve into family relationships in a useful way, they could look at what happens to families when a child gets either ME/CFS or another similarly debilitating chronic illness. i.e. rates of marriage breakdown and decreased extended family...

Effect of disease duration in a randomized Phase III trial of rintatolimod, an immune modulator for [ME/CFS], 2020, Mitchell et al

One of the criteria to be included in the 'Target Population' was 'PEM lasting more than 24 hours'. The participants in the non-target population presumably either didn't have PEM or had PEM that lasted less than 24 hours. It's not clear how the PEM criteria was determined, but it looks as...

Effect of disease duration in a randomized Phase III trial of rintatolimod, an immune modulator for [ME/CFS], 2020, Mitchell et al

Here are the scatter plots so you can make up your own mind. The first one is for the so-called 'target population', the second one is for everyone else. Illness duration is on the x axis, with different scales for each chart. % change in the time on the treadmill (from baseline to end of...

Effect of disease duration in a randomized Phase III trial of rintatolimod, an immune modulator for [ME/CFS], 2020, Mitchell et al

Answer #2 in how to market results from a blinded placebo controlled trial: focus on something entirely different from the planned outcomes. Here are the main results, comparing improvements in cohorts with the treatment and a placebo: So, it looks as though more patients treated with...

Effect of disease duration in a randomized Phase III trial of rintatolimod, an immune modulator for [ME/CFS], 2020, Mitchell et al

Sorry for my skepticism, but this looks like a lesson in 'how to get around the problems created by a blinded placebo controlled trial'. Answer 1: 'very detailed post hoc stratification' The measure, exercise treadmill tolerance, as measured once at baseline and once at trial end seems less...

Substrate utilisation of cultured skeletal muscle cells in patients with CFS, Tomas et al, 2020

Thanks @DMissa, that does help a lot. So, Complex V is making ATP and using oxygen to do it. When you stop Complex V from making ATP with the inhibitor, the use of oxygen should drop. How much it drops is an indication of how much ATP the Complex V was making before you inhibited it. Like...

Substrate utilisation of cultured skeletal muscle cells in patients with CFS, Tomas et al, 2020

CFS sample: 8 females, 1 male control sample: 9 females, 2 males I wondered about this, but the y axis labels (OCR pmol/min/ug protein) are exactly the same for the charts that are supposed to show basal respiration e.g 2a as the ones supposed to show ATP production e.g. 2c.

Substrate utilisation of cultured skeletal muscle cells in patients with CFS, Tomas et al, 2020

I think the figure references might be wrong here? Should it be "2A, E" as per the charts in my post above? Chart 2C is actually of ATP production. Using glucose as a substrate: Figure 2c is supposed to to be of ATP production. I don't understand the y-axis unit. Is that an error? How is...

Substrate utilisation of cultured skeletal muscle cells in patients with CFS, Tomas et al, 2020

The paper is open access. 9 CFS patients, 10 controls Glucose as a substrate for skeletal muscle cells A. Basal respiration - note significance between healthy controls and CFS - **. Pretty good. OCR is oxygen consumption rate. E. Maximal respiration - The cells were stressed somehow and...

Webinar: Scientific Research on ME/CFS in the Age of COVID-19 - Hanson, Lipkin, Nath - Oct 26th, 2020, 6 to 7pm ET.

Yes, I guess the oddness I mentioned was that Maureen had tried really hard to get across the idea of PEM to any clinicians listening. And Ian Lipkin didn't put his experience into context with that, at least I don't think he did. It would have been good if he had said, not all post-covid...

Nath He focussed largely on post-Covid19 neurological impacts. He noted that all human coronaviruses cause neurological complications. Showed images showing effectively brain damage, microhaemorrhages, macrophage infiltration into brain tissue and quoted a recent study that found that altered...

Webinar: Scientific Research on ME/CFS in the Age of COVID-19 - Hanson, Lipkin, Nath - Oct 26th, 2020, 6 to 7pm ET.

Lipkin He had a slide showing that 11% develop ME/CFS from EBV, Ross River fever and Q fever; 50% develop it after SARS and MERS, but none developed after some other infections. He threw XMRV in there, which seemed a bit confusing, but helped to make his point of doing work in the field for a...

Webinar: Scientific Research on ME/CFS in the Age of COVID-19 - Hanson, Lipkin, Nath - Oct 26th, 2020, 6 to 7pm ET.

I've just listened to it. It seemed more targeted at clinicians, and so there wasn't a lot that would be new to most of us here. There was emphasis on Me/CFS being a physical illness, with the material presented being quite convincing on that. It also had a wide scope, so the presentations...

Coronavirus - worldwide spread and control

In New Zealand, basically the only cases we have are those travellers coming from overseas, who become sick while in quarantine for two weeks in managed isolation facilities. Life is pretty much normal, except for any international travel requiring two weeks in quarantine upon return, and for...

    Restless legs syndrome

Moved from this thread I have just realised I have restless legs syndrome. I had had the vague idea that RLS involved legs spasming and so thought it was different to what I have. Normally, with just me to see my moving legs, I didn't think about it much. But I was driving my daughter crazy...
