Review Role of Ectopic Olfactory Receptors in the Regulation of the Cardiovascular–Kidney–Metabolic Axis, 2024, Beito et al

Role of Ectopic Olfactory Receptors in the Regulation of the Cardiovascular–Kidney–Metabolic Axis Abstract Olfactory receptors (ORs) represent one of the largest yet least investigated families of G protein-coupled receptors in mammals. While initially believed to be functionally restricted...

Human Olfactory Receptors: Novel Cellular Functions Outside of the Nose, 2018, Maßberg et al

I became interested in olfactory receptors as a couple of related genes (OR5V1 OR5AC2) cropped up in the PrecisionLife candidate genes and were flagged in one of the clusters in the analysis I was doing. It may not be significant, but it’s interesting. They are used for far more than smelling…...

Human Olfactory Receptors: Novel Cellular Functions Outside of the Nose, 2018, Maßberg et al

Human Olfactory Receptors: Novel Cellular Functions Outside of the Nose Maßberg, Désirée; Hatt, Hanns Abstract Olfactory receptors (ORs) are not exclusively expressed in the olfactory sensory neurons; they are also observed outside of the olfactory system in all other human tissues tested to...

Why is showering so PEM/OI inducing

It just occurred to me there’s probably a lot of sensory things going on when showering. A number of people have mentioned the elements. There’s thinking, coordination, a body having lots of touch signals, temperature changes, balance, noise, even movement/visual things. Looking at this from a...

Streaming nose first thing in the morning - allergies? Bug? Some weird ME/CFS thing?

No idea I’m afraid @Sasha but I have periods where my nose just starts running. Sometimes when I get up first thing, sometimes after eating, or just around the same time in the afternoon. No idea what causes these strange patches. Similarly with eyes watering (although other times they’re dry)...

Keeping clean when severe - a thread for tips

Yeah, my feet are grim atm. But when I am up to lying on the sofabed I use the bin from an old paper shredder, partially filled with water, with one of these in the bottom https://www.amazon.co.uk/dp/B004KX1OK8 and soak my feet in it and wiggle my toes a bit. Someone brings me this and after...

Keeping clean when severe - a thread for tips

The talk of showers reminded me I’d been thinking of sharing tips for keeping clean when you are unable to. It took me a long time to adapt, experiment snd learn, when at my best I manage to keep clean well, when less good it’s harder. I thought it would be useful for us to share approaches...

Why is showering so PEM/OI inducing

It’s been many years since I was able to but even in my early years of ME/CFS it would be a trigger. Even if sitting on the floor, the getting to the bathroom, washing, drying, getting back from the bathroom… I assumed the sustained activity and being upright was the reason, some other movements...

Review [+ Case Report] [POT] With [CF] After HPV Vaccination as Part of the “Autoimmune/Auto-inflammatory Syndrome Induced by Adjuvants”, 2014, Tomljenovic+

Yeah, acknowledging and understanding the rare reactions better (as well as increasing understanding of coincidences as @Jonathan Edwards has highlighted) would seem a sensible approach I think. Having significant changes in health and quality of life just dismissed doesn’t help anyone and only...

Review [+ Case Report] [POT] With [CF] After HPV Vaccination as Part of the “Autoimmune/Auto-inflammatory Syndrome Induced by Adjuvants”, 2014, Tomljenovic+

It’s an interesting thought, I’ve always thought adjuvants were pretty benign. As mentioned before I had terrible reactions to both AZ and Pfizr covid vaccines. I’d presumed something interesting about the spike proteins and my body, but maybe there’s some weird immune overreaction to adjuvants...

    Running FLAMES on DecodeME data

Glutamate receptors came up in some analysis of the Zhang paper and also seem prominent in my clustering analysis of PrecisionLife’s candidate genes from DecodeME data. I guess we’re looking at the same genes and gene set databases but still, may be interesting.

Personal project: Sick Genes, a website for compiling significant gene findings from studies on ME/CFS and other conditions

Great to see you find a way to get back to this @forestglip and thank you for all the thought and work you’ve put in. And for sharing your thinking. It’s always such a difficult balance to reach isn’t it? I was thinking similarly on my recent projects and used to see the same dilemma often with...

Noise cancelling headphones! Oh Joy! (and other ways to block sound)

A pretty good way of getting replacement pads for headphones, if you’re okay with it and know exactly what you want, is aliexpress. I was replacing some cushions in an old pair of AKG headphones and originally found some in the UK, but then found and bought basically the same for a fraction of...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

Apologies for the long posts but it seemed important to outline the process and I’ve been hesitant to share any biological data until the context had been explained in case it was all rubbish! Some configuration details and then the results: The source data from GTEx (I use v10 for this...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

A continuation on my above posts. I’ll try to explain a bit of the process I went through and the theory then how I decided to look at the number clusters that I did. In clustering theory, there's both the silhouette (a measure of cohesion, or how similar a data point is to other data points in...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

Yeah. It’s one of the things that is an option in my scripts. There are quite a few options for exploring the data in different ways (mean or median, log transformation or not, z-score or not). This wasn’t clever design. It was the result of an iterative process of failures, of exploration and...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

Some charts of clustering as a precursor to some more explanation. These show the relationship between the number of clusters (k) and two statistical measures: the Sum of Squared Errors (SSE) and the Silhouette Coefficient. Both are for the same underlying data, one without z-score normalisation...

Does ME/CFS affect facial expressions and voice with negative consequences on social function?

Facial expression, not like those disorders, but obviously those who know me can tell how I am. But that’s very different. Voice there is a more clear impact on, difficult to describe but it sort of ‘goes’ or becomes a bit off (or weak as @StellariaGraminea says) so I think something muscular...

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

Thanks Simon! In all honesty I don’t know. I’m hoping people with a background and better understanding in all this can tell me. My scripts do allow analysis of mean or median tpm values and with or without z-score normalisation, so people could try different approaches if more appropriate.

Preprint Identification of Novel Reproducible Combinatorial Genetic Risk Factors for [ME] in [DecodeME Cohort] and Commonalities with [LC], 2025, Sardell+

A longer but hopefully readable explanation… An approach that I have seen used is looking at expression of genes in tissues, finding genes which are more expressed together in particular tissues. This seems to often be linked in terms of mechanisms, or so I’ve read. So I took the gene list and...
