    Genetics: Chromosome 6 BTN2A2 and BTN3A3 (BTN2A1)

Did anyone ever contact these Cambridge BTN researchers? I know we talked about it but I can't remember what came of it. @Jonathan Edwards

Preprint Biological Insights from Genome-Wide Association Studies and Whole Genome Sequencing of [ME/CFS], 2026, Maccallini et al

I do wonder if there are any long covid (or MECFS) papers that we have dismissed on the basis of describing unusual cytokine activity as 'inflammation' or talking about 'T cell exhaustion' or whatever that might be telling us something important underneath the usual memes.

    Eccentric medium spiny neuron (eMSN)

Edit: Just quoted this post earlier to save energy but the relevant part to what was posted above is that the forkhead box protein (FOXP) transcriptional network is apparantly induced by IFNy, amongst other 'inflammatory' signals.

Identification of a forkhead box protein transcriptional network induced in human neutrophils in response to inflammatory stimuli, 2023, Ismailova+

So in laymans terms is it possible there's a story here that could go something like t cells> ifng> foxp4>foxp1>eMSNs Or @hotblack 's memory NK cells or similar on the immune end? I have just discovered NK cells produce IFNy too (probably not news to most of you!) It's all a bit six degrees...

Obviously what you say here is hypothetical but do you mean it in the sense of a massive finding being discovered 'last week' but not written up/published by the lab yet, so we are all unaware? That is an exciting idea!

The genetic architecture of fibromyalgia across 2.5 million individuals, 2025, Kerrebijnet al.

Since there are drugs targets suggested here including a repurposing opportunity, it seems like it's very important to figure out if these two genes do form an important pathway in MECFS. Are there any research groups that it might be of interest to? Tagging @James Cox just incase it interests...

Interleukin 6 as a Treatment Target for Depression: A Proof-of-Concept Randomized Clinical Trial, Foley et al., 2026

This just sums up everything wrong with modern science and science reporting. This was a negative result the team are spinning as possibly positive in order to keep their funding. The media is reporting on it like it's real, so the public will believe the treatment is effective. Really...

Preprint Monocyte Oxidative Stress Underlies Persistent Immune Activation in Long-COVID Postural Orthostatic Tachycardia Syndrome, 2026, Abd-Eldayem et al.

It seems that we've got several papers inc Jackie Cliffs suggesting links to TEMRA T Cells, in two of them secreting IFNy (in the Cliff paper IFNy secretion - and CD38 incidentally - was only seen in severe patients irrc correctly - here it is correlated with severity) . Obviously this LC paper...

    Eccentric medium spiny neuron (eMSN)

So from the genetic data we have, this is the type of neuron that is likely to be causing MECFS pathology? If thats the case, obviously that could be as part of an immune/brain loop or a brain only loop. And we don't know the specific dynamics of the pathology. But just in terms of drugs that...

UK: Financial Times: NHS to trial GPs no longer handing out ‘sick notes’

The idea of rest as being beneficial is being memory holed, along with ideas like 'people should be able to afford to feed their kids' and 'catching COVID-19 is something you should probably avoid'.

UK: Financial Times: NHS to trial GPs no longer handing out ‘sick notes’

Well this guarantees people with new onset MECFS and MECFS type long covid will have worse outcomes and worse levels of disablity. Never forget that PACE was sold as a way of getting people off benefits and back to work. This is all so sinister. The way everything is going it's like a...

    The optic nerve in ME/CFS

Yes and why I am desperately hoping those immune linked genes pan out and we're not left with a brain only signalling loop.

    The optic nerve in ME/CFS

The more I learn about how little we understand the brain, the angrier I get at the doctors and psychologists who were so confident that this symptom was just a manifestation of anxiety.

I have experienced this to a greater or lesser degree ever since my prodromal onset.

    Broken Battery Updates

It's one of the worst possible ways to describe the illness! It leads to so much stigma and confusion. I felt ill when I was mild. Like I always had a cold or was getting over one. And even when I was so exhausted I would have to rest in bed I primarily felt really f-king ill! So it took me...
