Genetics: Chromosome 6 FBXL4, POU3F2/BRN2

Found this preprint:

There's a lot in this paper but one thing caught my eye in Fig. 6 where they were doing some functional studies with POU3F2 overexpression.

The background for this is that cultured neurons will usually exhibit some spontaneous firing even without any external stimulation (the right column is with electrical stimulation, the left is without).

When you co-culture neurons and astrocytes, this causes periodic "network bursts" of coordinated electrical activity every ~60 mins across the plate without any electrical stimulation (that's what sgNT2+ast is showing in the bottom left).

Turns out that if you strongly induce POU3F2 expression in neurons, without astrocytes and without electrical stimulation, they start firing in coordinated bursts every 15 mins.

So this gives us something that POU3F2 can possibly do outside of early neural development, which might be the more fruitful route given ME/CFS is adult onset. The interesting question is what these variants alter POU3F2 induction or suppression in response to.

That's the cells taking their masks off and removing their earplugs and saying "Has that guy in the white coat gone yet. FFS?" Like the octopuses crawling out of their tanks when the zoologists go home to bed.
