It also goes on to state that Notch signalling has a role in neuronal function and development (among many other roles).The Notch signaling pathway is important for cell-cell communication, which involves gene regulation mechanisms that control multiple cell differentiation processes during embryonic and adult life.
i checked my WGS and am heterozygous for a missense mutation on NOTCH1, the highest ranked gene on your list. Clinvar lists the variant as benign, and CADD = 22.5 which seems high.
I also have a 2nd heterozygous intron variant listed as significant on NOTCH1.
